ABSTRACT

Objective:To investigate the taxonomic structure and diversity of endophytic fungi from Datura metel and screen the strains with anti-dermatophyte activities, so as to provide resources for the development of new lead compounds against dermatomycosis. Method:Endophytic fungi were isolated from the roots, stems and leaves of D. metel after tissue block incubation and then identified by morphological analysis and rDNA-internal transcribed spacer(ITS) sequencing. Their anti-dermatophyte activities were detected by agar diffusion assay. Result:A total of 292 strains of endophytic fungi were isolated from D. metel, belonging to 34 genera, with Fusarium (72.97%) in roots, Fusarium (37.25%) and Plectosphaerella (28.43%) in stems, and Colletotrichum (39.66%) in leaves as the dominant species. The isolation rate (89.23%), colonization rate (84.62%), and diversity index (1.82) of endophytic fungi in leaves were significantly higher than those in roots (70.48%, 70.48% and 1.23) and stems (69.39%,68.03% and 1.64). The determination of anti-dermatophyte activities of 35 endophytic fungal fermented filtrates showed that the strains exhibiting inhibitory activities against Microsporum canis, Trichosporon mucoides, Trichophyton rubrum and Candida albicans accounted for 97.14%, 71.43%, 45.71%, and 25.71%, respectively. Among them, six strains (17.14%), namely Fusarium sp. R1, Penicillium sp. R5, Aspergillus sp. R7, Metarhizium sp. S18, Diaporthe sp. S19, and Glomerella sp. L57, all inhibited the four types of cutaneous fungal pathogens. Conclusion:The endophytic fungi in D. metel are diverse, and the proportion of endophytic fungi possessing anti-dermatophyte activities is high, allowing them to serve as potential resources for the development of new anti-dermatophyte agents.

ABSTRACT

Ovarian cancer has the characteristics of difficulty in early diagnosis, easy recurrence and resistance, so its mortality rate ranks first in gynecological tumors. Existing treatment methods such as surgery, radiotherapy and chemotherapy are not very effective, thus immunotherapy has emerged. Immune checkpoint programmed death-1 (PD-1) and its ligand PD-L1 can inhibit T cell proliferation and mediate tumor immune escape. At present, PD-1/PD-L1 inhibitors have made breakthrough progress in a variety of solid tumors, and they have achieved initial efficacy in clinical treatment of ovarian cancer.

ABSTRACT

Increasingly, researches have shown that long non-coding RNA (lncRNA) plays an important role in the oncogenesis and development of various tumors. Small nucleoli RNA host gene 3 (SNHG3) is a newly discovered lncRNA whose abnormally high expression is closely related to the overall survival and prognosis of tumor patients. SNHG3 can regulate the oncogenesis and development of tumors by endogenous competitive adsorption of miRNA, regulating cell cycle, mediating epithelial and mesenchymal transformation, and activating multiple signaling pathways. Therefore, in-depth research on the carcinogenesis mechanism of SNHG3 is helpful for early diagnosis, targeted therapy and prognostic assessment of relevant tumors. This paper reviews latest research progress on the expression and mechanism of SNHG3 in breast cancer, ovarian cancer, kidney cancer, liver cancer, stomach cancer, colon cancer, osteosarcoma and head and neck tumors to provide references for future studies.

ABSTRACT

In this study, we proposed an approach to interpret the classification of body constitution based on the Japanese Version of Constitution in Chinese Medicine Questionnaire (CCMQ-J) in terms of an augmented questionnaire combining seven independent questionnaires. The augmented questionnaire consists of 254 questions in terms of seven categories of attributes, which are the (i) basic information (BI), (ii) disease (DI), (iii) social factors (SO), (iv) mental factors (ME), (v) dietary habits (DH), (vi) sleeping state (SL), and (vii) sub-health (SH). The partial least square (PLS) regression has been adopted to model the correlations between the scores of body constitutions and the questions, and their results show that the body constitution can be represented by the linear combination of the questions substantially (correlation coefficients between the true and predicted constitutions are all above 0.7). Moreover, by using the crowdsourcing technique in data collection, a total of 851 samples (350 males and 501 females between 20 and 85 years old) samples with diverse geographical backgrounds in Japan have been collected, from which new medical implications have been extracted through the discussion in a Traditional Chinese Medicine standpoint. This study serves as a crucial step for validating the philosophy of ancient Chinese medicine by the state-of-the-art information science techniques and facilitating the use of the CCMQ-J in public healthcare.

ABSTRACT

Hypotension is one of the potential causes of dizziness. In this review, we summarize the studies published in recent years about the electrophysiological and pharmacological mechanisms of hypotension-induced dizziness and the role of the vestibular system in the control of blood pressure in response to hypotension. It is postulated that ischemic excitation of the peripheral vestibular hair cells as a result of a reduction in blood flow to the inner ear following hypotension leads to excitation of the central vestibular nuclei, which in turn may produce dizziness after hypotension. In addition, excitation of the vestibular nuclei following hypotension elicits the vestibulosympathetic reflex, and the reflex then regulates blood pressure by a dual-control (neurogenic and humoral control) mechanism. In fact, recent studies have shown that peripheral vestibular receptors play a role in the control of blood pressure through neural reflex pathways. This review illustrates the dual-control mechanism of peripheral vestibular receptors in the regulation of blood pressure following hypotension.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the intervention effect of Shugan Jianpi Formula (, SGJPF) on a breast cancer mouse model with depression and investigate the underlying mechanism of SGJPF in preventing the development of breast cancer.</p><p><b>METHODS</b>The breast cancer model was induced by inoculation of breast cancer cells, the depression model was induced by chronic stress stimuli, and the depression cancer model was established by combining the two factors. The mice were divided into 7 groups: normal control, depression model, tumor model, depression tumor model, SGJPF, chemotherapy, and SGJPF+chemotherapy groups. The last 3 groups were depression breast cancer mice and treated respectively with SGJPF, chemotherapy drug gemcitabine (GEM), and SGJPF alongside GEM. The condition of the mice was evaluated by the expression of 5-hydroxytryptamine in hippocampus after the sucrose water test and open field test, weight change, and survival time. Tumor growth was monitored with in vivo imaging. Flow cytometry was used to analyze the level of myeloid-derived suppression cell (MDSC) in the mouse spleen, T cell subsets, and the early apoptosis of CD8T cells.</p><p><b>RESULTS</b>The SGJPF+GEM group had the highest inhibition rate and the longest survival time (P<0.01). The MDSC level and the apoptosis rate of CD8T cells was the highest in the SGJPF+GEM group (P<0.05).</p><p><b>CONCLUSIONS</b>Depressive disorders and tumor growth could suppress the immune function of mice to different degrees, and the microenvironment in late 4T1 inflammatory breast cancer may play an important role in the pathological process. SGJYF could regulate the immune microenvironment by reducing CD8T lymphocyte apoptosis and tumor cell activity, increasing immune surveillance capability, and inhibiting MDSC proliferation, thus prolonging the survival time of tumor-bearing mice.</p>

ABSTRACT

<p><b>OBJECTIVE</b>Aging is associated with a progressive decline in muscle strength, muscle mass and impaired physical function, which reduces mobility and impairs quality of life in the elderly population. The 6-12 months of exercise can enhance the muscle strength, but these improvements can only be maintained for a short period. In this study, we investigated the effects of long-term Tai Chi (TC) exercise on muscle strength of lower extremities.</p><p><b>METHODS</b>A cross-sectional study was conducted in 205 long-term TC practitioners (age: 60-89 years) and 205 age and gentle matched controls who did not practice TC. Each of the activity group was further divided into three distinct age groups: G1, 60-69 years; G2, 70-79 years; and G3, 80-89 years. Hand-held dynamometery was used to measure the maximum isometric strength of iliopsoas, quadriceps femoris, tibialis anterior and hamstrings in both sides of the participants. Unpaired t tests were performed to compare the difference of strength between the TC and non-Tai Chi (NTC) groups. Multivariate analysis of variance (MANOVA) was used to compare the lower muscle strengths among the different age groups in the TC and NTC groups. Pearson's correlations were used to quantify the linear relationship between the months of TC practice and lower limbs muscle strength.</p><p><b>RESULTS</b>The inter-rater reliabilities of iliopsoas, quadriceps femoris, tibialis anterior and hamstrings were intraclass correlation coeffificient (ICC) (1,1) = 0.895 (0.862-0.920), ICC (2,2) = 0.905 (0.874-0.928), ICC (3,3) = 0.922 (0.898-0.941) and ICC (4,4) = 0.930 (0.908-0.947). The strength of the muscles in the TC group did not differ among different age groups (P>0.05). The strength of iliopsoas, quadriceps femoris, tibialis anterior and hamstrings in TC group was higher than that in the NTC group (P<0.05). A correlation between muscle strength and extension of the exercise period was positive (P<0.05).</p><p><b>CONCLUSIONS</b>Results shed light on the orientation and magnitude of long-term TC in preventing muscle strength loss with aging. TC might be a good form to slow down the trend of age-related decline in muscle strength in community-dwelling population.</p>
